This protein is an important cell cycle checkpoint kinase that phosphorylates; thus, it functions as a regulator of a wide variety of downstream proteins, including tumor suppressor proteins p53 and BRCA1, checkpoint kinase CHK2, checkpoint proteins RAD17 and RAD9, and DNA repair protein NBS1. This protein and the closely related kinase ATR are thought to be master controllers of cell cycle checkpoint signaling pathways that are required for cell response to DNA damage and for genome stability. Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant was absent in 251078 control chromosomes (gnomAD). c.7010_7011delGT has been reported in the literature in multiple individuals affected with Ataxia-Telangiectasia (Telatar_1996, Telatar_1998, Li_2000, Buzin_2003, Podralska_2014). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Three ClinVar submitters (evaluation after 2014) cite the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -

This mutation has been well documented in the literature in a compound heterozygous state in multiple individuals with Ataxia Telangiectasia (A-T) (Buzin CH et al. Hum.
