This variant was reported in individual(s) with features consistent with neurofibromatosis type 1 (Upadhyaya M et al. Hum Genet, 1997 Jan;99:88-92; Fahsold R et al. Am J Hum Genet, 2000 Mar;66:790-818; Ars E et al. J Med Genet, 2003 Jun;40:e82; Griffiths S et al. Fam Cancer, 2007;6:21-34; Pros E et al. Hum Mutat, 2008 Sep;29:E173-93; Ambry internal data). This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site and will result in the creation or strengthening of a novel splice acceptor site.
