The glutamic acid at codon 134 is replaced by valine, an amino acid with dissimilar properties. This variant has been detected in trans with a MUTYH pathogenic mutation in an individual with multiple adenomatous polyps (Ambry internal data). Based on an internal structural analysis, this alteration eliminates a functionally critical interaction in base excision repair responsible for identifying and removing adenine from the substrate (Lee S et al. Proc. Natl. Acad. Sci. U.S.A., 2009 Nov;106:18497-502; Luncsford PJ et al. J. Mol. Biol., 2010 Oct;403:351-70).
