This missense variant replaces glutamic acid with glutamine at codon 170 of the MYL3 protein. Computational prediction tools indicate that this variant has a deleterious impact on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in three homozygous and three heterozygous individuals affected with hypertrophic cardiomyopathy (PMID: 37431535). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

The p.E170Q variant (also known as c.508G>C), located in coding exon 5 of the MYL3 gene, results from a G to C substitution at nucleotide position 508. The glutamic acid at codon 170 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
