Truncations downstream of this position have been classified as pathogenic by our laboratory (c.2255_2256delAA, p.Lys752fsX12; c.2392C>T, p.Arg798X; c.2400C>G, p.Tyr800X). The variant was absent in 251042 control chromosomes (gnomAD). The variant c.2114_2118delAAGAA, has been reported in the literature in an individual affected with endometrial carcinoma (Ring_2016). This data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Four submitters have provided clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation.
