This sequence change replaces glycine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 312 of the TGFBR1 protein (p.Gly312Asp). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TGFBR1-related conditions. ClinVar contains an entry for this variant (Variation ID: 222837). This variant disrupts the p.Gly312 amino acid residue in TGFBR1. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 16799921, 19542084, 26848186, 26877057, 30739908). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing.
