The homotetramer enzyme catalyzes the initial step of the mitochondrial fatty acid beta-oxidation pathway. Defects in this gene cause medium-chain acyl-CoA dehydrogenase deficiency, a disease characterized by hepatic dysfunction, fasting hypoglycemia, and encephalopathy, which can result in infantile death. Experimental studies have shown that this missense change affects ACADM function (PMID: 19224950). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on ACADM protein function. ClinVar contains an entry for this variant (Variation ID: 226089). This variant is also known as R223G. This missense change has been observed in individual(s) with Medium-chain acyl-coenzyme A dehydrogenase deficiency and/or medium-chain acyl-coenzyme A dehydrogenase deficiency (PMID: 15832312).
