Desmogleins are calcium-binding transmembrane glycoprotein components of desmosomes, cell-cell junctions between epithelial, myocardial, and other cell types. The encoded preproprotein is proteolytically processed to generate the mature glycoprotein. This gene is present in a gene cluster with other desmoglein gene family members on chromosome 18. Methionine (Met) at position 757 is not conserved in evolution and 1 mammal (elephant) carries a threonine (Thr), raising the possibility that this change may be tolerated. In summary, while th e clinical significance of the p.Met757Thr variant is uncertain, the presence of the variant amino acid in other species suggests that it is more likely to be b enign. -
