Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.6e-05 in 246580 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.8585A>G has been reported in the literature as a non-informative genotype (second allele not specified or classified as a benign variant) in individuals affected with hearing loss (example, Garcia-Garcia_2013, Sloan-Heggen_2016). These report(s) do not provide unequivocal conclusions about association of the variant with Usher Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 23441107, 26969326, 35813073). Two submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. One submitter classified the variant as likely benign and one submitter classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. -
