The aspartic acid at codon 758 is replaced by asparagine, an amino acid with highly similar properties. This alteration was identified in a patient with endometrial cancer diagnosed at age 41 with a family history of colorectal cancer in their father at age 80 (Özdemir TR et al. Balkan Med J, 2019 01;36:37-42). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
