To our knowledge, functional assays have not been reported for this variant, however studies have shown that an SDHAF2 protein containing aa37-151 is defective for both SDHA binding and flavination, suggesting this variant would show similar defects (PMID: 32887801). This variant has been reported in an individual affected with a carotid body paraganglioma (PMID: 31687641, 33748650). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Based on the available evidence, this variant is classified as Likely Pathogenic. -

The c.347G>A (p.W116*) alteration, located in exon 3 (coding exon 3) of the SDHAF2 gene, consists of a G to A substitution at nucleotide position 347. This changes the amino acid from a tryptophan (W) to a stop codon at amino acid position 116. This alteration occurs at the 3' terminus of theSDHAF2 gene, is not expected to trigger nonsense-mediated mRNA decay, and only impacts the last 51 amino acids of the protein. However, premature stop codons are typically deleterious in nature and a significant portion of the protein is affected. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This alteration has been detected in a patient diagnosed with a paraganglioma at age 30 (Ambry internal data). This alteration is predicted to disrupt a conserved domain of the protein (Ambry internal data). Based on the available evidence, this alteration is classified as likely pathogenic. -
