The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.6450A>C in individuals affected with Hereditary Breast and Ovarian Cancer and no experimental evidence demonstrating its impact on protein function have been reported. Co-occurrence with one other pathogenic variant has been reported in our database (BRCA2 c.(7007+1_7008-1)_(7805+1_7806-1)del, p.Thr2337_Arg2602del), providing supporting evidence for a benign role. Two ClinVar submissions from other clinical diagnostic laboratories (evaluation after 2014) cites the variant as uncertain significance.
