Neurofibromatosis, type 1 Pathogenic:2
Studies have shown that disruption of this splice site results in skipping of exon 38, also referred to as exon 30, and introduces a premature termination codon (PMID: 18546366; Invitae). The resulting mRNA is expected to undergo nonsense-mediated decay. ClinVar contains an entry for this variant (Variation ID: 230938). Disruption of this splice site has been observed in individual(s) with clinical features of neurofibromatosis, type 1 (PMID: 18546366, 31717729, 31730495). This variant is not present in population databases (gnomAD no frequency). This sequence change affects a donor splice site in intron 38 of the NF1 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. -

Hereditary cancer-predisposing syndrome Pathogenic:1
The c.5812+1G>A intronic pathogenic mutation (also known as c.5749+1G>A) results from a G to A substitution one nucleotide after coding exon 39 of the NF1 gene. This mutation was identified in a NF1 patient and mRNA analysis indicated that it leads to skipping of coding exon 39 (referred to as exon 30), which is predicted to cause aframe shift and premature stop codon (Pros, E et al. Hum Mutat. 2008 Sep;29(9):E173-93). In addition to the clinical data presented in the literature, since alterations that disrupt the canonical splice donor site are typically deleterious in nature, this alteration is interpreted as a disease-causing mutation (ACMG Recommendations for Standards for Interpretation and Reporting of Sequence Variations. Revision 2007. Genet Med. 2008;10:294). -
