The c.18_19delACinsTG variant, located in coding exon 1 of the MRE11A gene, results from an in-frame deletion of AC and insertion of TG at nucleotide positions 18 to 19. This results in the substitution of the leucine residue for a valine residue at codon 7, an amino acid with highly similar properties. This amino acid position is highly conserved on limited alignment. In silico splice site analysis predicts that this alteration may weaken the native splice donor site. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
