In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces valine with isoleucine at codon 1038 of the ATM protein (p.Val1038Ile). The valine residue is highly conserved and there is a small physicochemical difference between valine and isoleucine. -

The p.V1038I variant (also known as c.3112G>A), located in coding exon 20 of the ATM gene, results from a G to A substitution at nucleotide position 3112. The valine at codon 1038 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
