The p.Y433* pathogenic mutation (also known as c.1299T>A) located in coding exon 12 of the NF1 gene, results from a T to A substitution at nucleotide position 1299. This changes the amino acid from a tyrosine to a stop codon. This mutation was identified in a 2 yo boy with features of NF1 (WimmerK et al,Pediatr. Res. 2005 Dec; 58(6):1265-8).<span style="font-family:arial,sans-serif; font-size:9pt">In addition to the clinical data presented in the <span style="font-family:arial,sans-serif; font-size:9pt">literature, since premature stop codons are typically deleterious in nature, this alteration is interpreted as a disease-causing mutation (ACMG Recommendations for Standards for Interpretation and Reporting of Sequence Variations.
