The c.4801A>G variant (also known as p.S1601G), located in coding exon 31 of the ATM gene, results from an A to G substitution at nucleotide position 4801. The serine at codon 1601 is replaced by glycine, an amino acid with similar properties. This variant has been identified in conjunction with a pathogenic ATM mutation in an individual with a diagnosis of ataxia-telangiectasia (A-T) (Kim J et al. Nature, 2023 Jul;619:828-836). In silico splice site analysis predicts that this alteration will result in the creation or strengthening of a novel splice acceptor site; however, direct evidence is insufficient at this time (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear. -
This missense variant replaces serine with glycine at codon 1601 of the ATM protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in one individual affected with breast cancer (PMID: 28779002). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
