The p.N112I variant (also known as c.335A>T), located in coding exon 2 of the CHEK2 gene, results from an A to T substitution at nucleotide position 335. The asparagine at codon 112 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
