The arginine at codon 87 is replaced by leucine, an amino acid with dissimilar properties. In one functional study, this alteration showed loss of inhibition of cell cycle arrest (Yarbrough WG et al. J Natl Cancer Inst, 1999 Sep;91:1569-74). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. - |
