Variant summary: NOTCH1 c.17_18delinsTT (p.Ala6Val) is part of a multinucleotide combination of 9-136545770-G-A )c.17C>T; p.Ala6Val) and 9-136545769-C-A (c.18G>T, p.Ala6=) that results in a non-conservative amino acid change in the encoded protein sequence. Two of four in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 8e-05 in 1359860 control chromosomes in the gnomAD database, including 1 homozygotes. The observed variant frequency is approximately 128.25 fold of the estimated maximal expected allele frequency for a pathogenic variant in NOTCH1 causing Adams-Oliver Syndrome 5 phenotype (6.3e-07). To our knowledge, no occurrence of c.17_18delinsTT in individuals affected with Adams-Oliver Syndrome 5 and no experimental evidence demonstrating its impact on protein function have been reported.
