For these reasons, this variant has been classified as Pathogenic. Studies have shown that this variant is associated with skipping of exons 7 and 8 but is expected to preserve the integrity of the reading frame (PMID: 9973282). Disruption of this splice site has been observed in individual(s) with Pfeiffer syndrome (PMID: 9973282, 10394936, 12884424, 27481450, 9048930). This variant is also known as c.1119-1G>T in the literature. This variant is not present in population databases (ExAC no frequency). This sequence change affects an acceptor splice site in intron 7 of the FGFR2 gene. RNA analysis indicates that this variant induces altered splicing and likely results in a shortened protein product. -
