This results in the duplication of an extra residue between codons 562 and 563. This variant has been identified in conjunction with other LDLR variant(s) in individual(s) with features consistent with homozygous familial hypercholesterolemia (Reijman MD et al. Curr Opin Lipidol, 2023 Dec;34:287-295). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear. -
