The c.2068delC pathogenic mutation, located in coding exon 14 of the LDLR gene, results from a deletion of one nucleotide at nucleotide position 2068, causing a translational frameshift with a predicted alternate stop codon (p.H690Tfs*19). This alteration has been previously reported in multiple individuals in a familial hypercholesterolemia cohort (Tichý L et al. Atherosclerosis. 2012;223:401-8). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
Familial hypercholesterolemia Pathogenic:1
This sequence change creates a premature translational stop signal (p.His690Thrfs*19) in the LDLR gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in LDLR are known to be pathogenic (PMID: 20809525, 28645073). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with familial hypercholesterolemia (PMID: 11754108). This variant is also known as FsH669.
