CLCN4 (HGNC:2022): (chloride voltage-gated channel 4) The CLCN family of voltage-dependent chloride channel genes comprises nine members (CLCN1-7, Ka and Kb) which demonstrate quite diverse functional characteristics while sharing significant sequence homology. Chloride channel 4 has an evolutionary conserved CpG island and is conserved in both mouse and hamster. This gene is mapped in close proximity to APXL (Apical protein Xenopus laevis-like) and OA1 (Ocular albinism type I), which are both located on the human X chromosome at band p22.3. The physiological role of chloride channel 4 remains unknown but may contribute to the pathogenesis of neuronal disorders. This sequence change replaces valine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 275 of the CLCN4 protein (p.Val275Met). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with CLCN4-related conditions (PMID: 27550844, 33880059, 36385166). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 209117). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt CLCN4 protein function with a positive predictive value of 95%.
