This protein forms a complex with other SMAD proteins and binds DNA, functioning both as a transcription factor and tumor suppressor. The p.L219F variant (also known as c.657G>C), located in coding exon 5 of the SMAD3 gene, results from a G to C substitution at nucleotide position 657.
