The c.2095_2096delCA pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a deletion of two nucleotides at nucleotide positions 2095 to 2096, causing a translational frameshift with a predicted alternate stop codon (p.Q699Vfs*8). This alteration has been identified in multiple individuals diagnosed with breast and/or ovarian cancer (Chan GHJ et al. Oncotarget, 2018 Jul;9:30649-30660; Dorling et al. N Engl J Med. 2021 02;384:428-439). This alteration was also identified in a large, worldwide study of BRCA1/2 mutation positive families (Rebbeck TR et al. Hum Mutat, 2018 05;39:593-620). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
