This variant has been reported in an individual affected with breast cancer with multiple relatives affected by breast or endometrial cancer (Color internal data), and has been identified in high-risk hereditary breast and ovarian cancer individuals (PMID: 27425403, 29907814). This variant has also been referred to as c.7986delG, p.Glu2665Lysfs in the literature (PMID: 27425403). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of BRCA2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic. -
