For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the BRCA2 protein in which other variant(s) (p.Leu2653Pro) have been determined to be pathogenic (PMID: 17924331, 19043619, 21990134, 22678057, 23108138). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Studies have shown that disruption of this splice site results in skipping of exon 17, but is expected to preserve the integrity of the reading-frame (PMID: 28339459, 31191615). ClinVar contains an entry for this variant (Variation ID: 267689). Disruption of this splice site has been observed in individual(s) with breast cancer (PMID: 31528241). This variant is not present in population databases (gnomAD no frequency). This sequence change affects a donor splice site in intron 17 of the BRCA2 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and likely results in a shortened protein product. -
