The BRCA2 c.9118-1G>A variant (rs886040950) is reported in the literature in at least one individual affected with ovarian cancer (Cardoso 2018). This variant is also reported in ClinVar (Variation ID: 267716), but is absent from the Genome Aggregation Database, indicating it is not a common polymorphism. This variant disrupts the canonical splice acceptor site of intron 23, which is likely to negatively impact gene function. Based on available information, this variant is considered to be pathogenic. It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change affects an acceptor splice site in intron 23 of the BRCA2 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584). -
