The DMD c.10126delC; p.Leu3376Ter variant (rs886043676) is reported in the literature in several individuals affected with DMD or BMD (Tuffery 1995, Cho 2017). This variant is reported as pathogenic in ClinVar (Variation ID: 287624). This variant is also absent from general population databases (Exome Variant Server, Genome Aggregation Database), indicating it is not a common polymorphism. This variant induces an early termination codon and is predicted to result in a truncated protein or mRNA subject to nonsense-mediated decay. Based on available information, this variant is considered to be pathogenic. References: Cho A et al. Duchenne muscular dystrophy Pathogenic:1
This sequence change creates a premature translational stop signal (p.Leu3376*) in the DMD gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in DMD are known to be pathogenic (PMID: 16770791, 25007885). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individuals with Duchenne or Becker muscular dystrophy (PMID: 7581396, 27593222). This variant is also known as 10334delC. ClinVar contains an entry for this variant (Variation ID: 287624). For these reasons, this variant has been classified as Pathogenic. -
