Inborn genetic diseases Uncertain:1
The c.950C>T (p.S317L) alteration is located in exon 16 (coding exon 15) of the NDRG1 gene. This alteration results from a C to T substitution at nucleotide position 950, causing the serine (S) at amino acid position 317 to be replaced by a leucine (L). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
Charcot-Marie-Tooth disease type 4D Uncertain:1
Charcot-Marie-Tooth disease type 4 Uncertain:1
This sequence change replaces serine with leucine at codon 317 of the NDRG1 protein (p.Ser317Leu). The serine residue is moderately conserved and there is a large physicochemical difference between serine and leucine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with NDRG1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
