The c.1192C>G (p.L398V) alteration is located in exon 3 (coding exon 3) of the NLRP3 gene. This alteration results from a C to G substitution at nucleotide position 1192, causing the leucine (L) at amino acid position 398 to be replaced by a valine (V). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.

Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with NLRP3-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change replaces leucine with valine at codon 398 of the NLRP3 protein (p.Leu398Val). The leucine residue is moderately conserved and there is a small physicochemical difference between leucine and valine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
