The p.F422S variant (also known as c.1265T>C), located in coding exon 2 of the RBM20 gene, results from a T to C substitution at nucleotide position 1265. The phenylalanine at codon 422 is replaced by serine, an amino acid with highly dissimilar properties. This alteration has been reported in a hypertrophic cardiomyopathy (HCM) cohort; however, clinical details were limited (Lopes LR et al. Heart, 2015 Feb;101:294-301). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
