The asparagine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. The p.D1815N variant (also known as c.5443G>A), located in coding exon 35 of the ATM gene, results from a G to A substitution at nucleotide position 5443. The aspartic acid at codon 1815 is replaced by asparagine, an amino acid with highly similar properties. This alteration was not observed in 53 unselected men with breast cancer and was observed with an allele frequency of 0.0001 in 12490 male controls of Japanese ancestry (Momozawa Y et al. Nat Commun, 2018 10;9:4083). This amino acid position is not well conserved in available vertebrate species, and asparagine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
