The p.Ile563Val variant in OTOGL has not been previously reported in individuals with hearing loss, but has been identified in 2/8732 African chromosomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadinstitute.org; dbSNP rs 942588475). Although this variant has been seen in the general population, its f requency is low enough to be consistent with a recessive carrier frequency. Comp utational prediction tools and conservation analysis suggest that the p.Ile563Va l variant may not impact the protein, though this information is not predictive enough to rule out pathogenicity. In summary, the clinical significance of the p .Ile563Val variant is uncertain. ACMG/AMP Criteria applied: BP4 (Richards 2015). -
