The p.C991W variant (also known as c.2973C>G), located in coding exon 23 of the POLD1 gene, results from a C to G substitution at nucleotide position 2973. The cysteine at codon 991 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
