This sequence change replaces histidine, which is basic and polar, with glutamine, which is neutral and polar, at codon 863 of the POLD1 protein (p.His863Gln). -

The p.H863Q variant (also known as c.2589C>G), located in coding exon 20 of the POLD1 gene, results from a C to G substitution at nucleotide position 2589. The histidine at codon 863 is replaced by glutamine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6495 samples (12990 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of p.H863Q remains unclear. -
