This missense variant (also known as p.Ile334Met in the mature protein) replaces isoleucine with methionine at codon 355 of the LDLR protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with hypercholesterolemia (PMID: 28379029). This variant has been identified in 2/251230 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

The p.I355M variant (also known as c.1065C>G), located in coding exon 8 of the LDLR gene, results from a C to G substitution at nucleotide position 1065. The isoleucine at codon 355 is replaced by methionine, an amino acid with highly similar properties. This variant was reported in individual(s) with features consistent hypercholesterolemia (Tichý L et al. Physiol Res, 2017 Apr;66:S47-S54). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
