This alteration results from a T to C substitution at nucleotide position 1556, causing the leucine (L) at amino acid position 519 to be replaced by a proline (P). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces leucine with proline at codon 519 of the FANCI protein (p.Leu519Pro). The leucine residue is highly conserved and there is a moderate physicochemical difference between leucine and proline. This variant is not present in population databases (ExAC no frequency).
