The c.1988G>T (p.R663L) alteration is located in exon 17 (coding exon 15) of the MFN2 gene. This alteration results from a G to T substitution at nucleotide position 1988, causing the arginine (R) at amino acid position 663 to be replaced by a leucine (L). Based on data from the Genome Aggregation Database (gnomAD), the MFN2 c.1988G>T alteration was observed in 0.001% (4/282,798) total alleles studied. This amino acid position is conserved in available vertebrate species, except for lizard. The alteration is predicted deleterious by in silico models:_x000D_ _x000D_ The p.R663L alteration is predicted to be possibly damaging by Polyphen and deleterious by SIFT in silico analyses. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Charcot-Marie-Tooth disease type 2 Uncertain:1
This sequence change replaces arginine, which is basic and polar, with leucine, which is neutral and non-polar, at codon 663 of the MFN2 protein (p.Arg663Leu).
