The p.R663H variant (also known as c.1988G>A), located in coding exon 15 of the MFN2 gene, results from a G to A substitution at nucleotide position 1988. The arginine at codon 663 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
Charcot-Marie-Tooth disease type 2 Uncertain:1
This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 663 of the MFN2 protein (p.Arg663His). This variant is present in population databases (rs766735605, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with MFN2-related conditions.
