Pathogenic, criteria provided, single submitter | clinical testing | Ambry Genetics | Aug 14, 2023 | The c.1608_1609dupGG (p.A537Gfs*37) alteration, located in exon 11 (coding exon 10) of the HNRNPR gene, consists of a duplication of GG at position 1608, causing a translational frameshift with a predicted alternate stop codon after 37 amino acids. This alteration occurs at the 3' terminus of the HNRNPR gene, is not expected to trigger nonsense-mediated mRNA decay, and only impacts the last 15.7% of the protein. However, premature stop codons are typically deleterious in nature, the impacted region is critical for protein function, and a significant portion of the protein is affected (Ambry internal data). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on the available evidence, this alteration is classified as pathogenic. - |
