Uncertain significance, criteria provided, single submitter | curation | Broad Center for Mendelian Genomics, Broad Institute of MIT and Harvard | Nov 25, 2024 | The heterozygous p.Leu182Pro variant in GGPS1 was identified by our study in the compound heterozygous state, along with another variant of uncertain significance in one individual with neuromuscular disease (PMID: 35869884). Trio exome analysis revealed that this variant was in trans with the other variant. The variant has not been previously reported in the literature in individuals with neuromuscular disease and has been identified in 0.0007% (8/1179774) of European (non-Finnish) chromosomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadinstitute.org). Computational prediction tools and conservation analyses suggest that this variant may impact the protein, though this information is not predictive enough to determine pathogenicity. The p.Leu182Pro variant is located in a region of GGPS1 that is essential to protein folding and stability, suggesting that this variant is in a functional domain and slightly supports pathogenicity (PMID: 35869884). The phenotype of an individual heterozygous for this variant is highly specific for GGPS1 related muscular dystrophy based on dystrophic muscle histology and ultrastructural evidence of autophagic material and large mitochondria consistent with disease (PMID: 35869884). In summary, while there is some suspicion for a pathogenic role, the clinical significance of this variant is uncertain.
