This alteration results from a T to C substitution at nucleotide position 326, causing the isoleucine (I) at amino acid position 109 to be replaced by a threonine (T). Based on data from gnomAD, the C allele has an overall frequency of <0.01% (3/247036) total alleles studied. This alteration has been reported in a patient with childhood epilepsy in combination with a second TRIT1 alteration. No additional clinical information was provided (Balciuniene, 2019). This amino acid position is not well conserved in available vertebrate species. The in silico prediction for this alteration is inconclusive. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. - |
