Genes affected
TECTA (HGNC:11720): (tectorin alpha) The tectorial membrane is an extracellular matrix of the inner ear that contacts the stereocilia bundles of specialized sensory hair cells. Sound induces movement of these hair cells relative to the tectorial membrane, deflects the stereocilia, and leads to fluctuations in hair-cell membrane potential, transducing sound into electrical signals. Alpha-tectorin is one of the major noncollagenous components of the tectorial membrane. Mutations in the TECTA gene have been shown to be responsible for autosomal dominant nonsyndromic hearing impairment and a recessive form of sensorineural pre-lingual non-syndromic deafness. Uncertain significance, criteria provided, single submitterclinical testingInvitaeAug 24, 2022In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The methionine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. ClinVar contains an entry for this variant (Variation ID: 504680). This variant has not been reported in the literature in individuals affected with TECTA-related conditions. This variant is present in population databases (rs372282487, gnomAD 0.008%). This sequence change replaces threonine, which is neutral and polar, with methionine, which is neutral and non-polar, at codon 2134 of the TECTA protein (p.Thr2134Met). -

Likely benign, criteria provided, single submitterclinical testingLaboratory for Molecular Medicine, Mass General Brigham Personalized MedicineJun 09, 2016p.Thr2134Met in exon 23 of TECTA: This variant is not expected to have clinical significance because the threonine (Thr) at position 2134 is not conserved in ma mmals or evolutionary distant species, with 10 mammals having a methionine (Met) at this position. It has been identified in 3/66304 European chromosomes by t he Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org). -
