The encoded protein is a transcriptional activator that is required for normal development of the central nervous system, chondrogenesis and maintenance of cardiac and skeletal muscle cells. This alteration results from a G to A substitution at nucleotide position 2084, causing the arginine (R) at amino acid position 695 to be replaced by a lysine (K). Based on data from gnomAD, the A allele has an overall frequency of 0.001% (3/282862) total alleles studied. The highest observed frequency was 0.002% (3/129164) of European (non-Finnish) alleles. This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. - |
