Genes affected
MYBPC3 (HGNC:7551): (myosin binding protein C3) MYBPC3 encodes the cardiac isoform of myosin-binding protein C. Myosin-binding protein C is a myosin-associated protein found in the cross-bridge-bearing zone (C region) of A bands in striated muscle. MYBPC3 is expressed exclusively in heart muscle and is a key regulator of cardiac contraction. Mutations in this gene are a frequent cause of familial hypertrophic cardiomyopathy. Hypertrophic cardiomyopathy Pathogenic:2
Pathogenic, criteria provided, single submitter | clinical testing | Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine | Sep 20, 2011 | The Trp1098X variant has been reported in one individual with HCM (Millat 2010) and has been identified in 1 proband with HCM tested by our laboratory. This va riant leads to a premature stop at codon 1098 and is predicted to lead to a trun cated or absent protein. Loss of function of the MYBPC3 gene is an established disease mechanism in HCM patients, which makes it highly likely that the Trp1098 X variant is pathogenic. - |
Pathogenic, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Mar 16, 2017 | For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in MYBPC3 are known to be pathogenic (PMID: 19574547). This variant has been reported in several individuals affected with hypertrophic cardiomyopathy (PMID: 25031304, 23233322, 20624503). This sequence change creates a premature translational stop signal at codon 1098 (p.Trp1098*) of the MYBPC3 gene. It is expected to result in an absent or disrupted protein product. - |
Hypertrophic cardiomyopathy 4;C3715165:Left ventricular noncompaction 10 Pathogenic:1
Pathogenic, criteria provided, single submitter | clinical testing | Center for Genomics, Ann and Robert H. Lurie Children's Hospital of Chicago | Mar 30, 2021 | MYBPC3 NM_000256.3 exon 29 p.Trp1098* (c.3293G>A): This variant has been reported in the literature in at least 7 individuals with hypertrophic cardiomyopathy (HCM) (Millat 2010 PMID:20624503, Harris 2011 PMID:21415409, Kassem 2012 PMID:23233322, Helms 2014 PMID:25031304). This variant is not present in large control databases but is present in ClinVar (Variation ID:42701). Evolutionary conservation and computational predictive tools for this variant are limited or unavailable. This variant creates a premature stop at this codon which results in an absent or abnormal protein. Loss of function variants are a known mechanism of disease for this gene (Marston 2009 PMID:19574547). In summary, this variant is classified as pathogenic based on the data above. - |
