This alteration results from an A to G substitution at nucleotide position 1003, causing the isoleucine (I) at amino acid position 335 to be replaced by a valine (V). Based on data from gnomAD, the G allele has an overall frequency of 0.003% (1/31368) total alleles studied. The highest observed frequency was 0.007% (1/15404) of European (non-Finnish) alleles. This nucleotide position is highly conserved in available vertebrate species. This missense alteration is located in a region that has a low rate of benign missense variation (Lek, 2016; Firth, 2009). The in silico prediction for this alteration is inconclusive. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
