To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The p.Gln3361Pro variant is reported with the allele frequency (0.003%) in the gnomAD Exomes and is novel (not in any individuals) in 1000 Genomes. The amino acid Gln at position 3361 is changed to a Pro changing protein sequence and it might alter its composition and physico-chemical properties. In silico tools predict the variant to be tolerated. The residue is conserved across species. The amino acid change p.Gln3361Pro in BRCA2 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. The variant allele was found at a frequency of 3.7e-05 in 273876 control chromosomes in the gnomAD database and in one study surveying a control Han Chinese population (Dong_2021), including 1 homozygote in the gnomAD database. This frequency is not significantly higher than expected for a pathogenic variant in BRCA2 causing Hereditary Breast And Ovarian Cancer Syndrome (3.7e-05 vs 0.00075), allowing no conclusion about variant significance. c.10082A>C has been reported in the literature in one individual affected with Hereditary Breast And Ovarian Cancer Syndrome (Dorling_2021), however this report does not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. The variant was identified in control databases in 9 of 245868 chromosomes (1 homozygous) at a frequency of 0.00004 (Genome Aggregation Database Feb 27, 2017). The variant was observed in the South Asian population in 9 of 30774 chromosomes (1 homozygous, freq: 0.0003); it was not observed in the African, Other, Latino, European, Ashkenazi Jewish, East Asian, or Finnish populations.
