This protein is one of six core proteins in the telosome/shelterin telomeric complex, which functions to maintain telomere length and to protect telomere ends. Through its interaction with other components, this protein plays a key role in the assembly and stabilization of this complex, and it mediates the access of telomerase to the telomere. Multiple transcript variants encoding different isoforms have been found for this gene. This gene, which is also referred to as TPP1, is distinct from the unrelated TPP1 gene on chromosome 11, which encodes tripeptidyl-peptidase I. [provided by RefSeq, Jul 2008]

This individual's healthy mother carried c.1213C>A in the heterozygous state and had normal telomere length. This variant has been reported in ClinVar (Variation ID: 208984) and is rare (<0.1%) in a large population dataset (gnomAD: 13/251316 total alleles; 0.005173%; no homozygotes). In vitro functional studies indicate that p.Pro405Thr may result in a modest decrease in binding between ACD and TIN2,however the clinical significance of this finding is uncertain. The proline residue at this position is evolutionarily conserved in most mammalian species assessed. We consider the clinical significance of ACD c.1213C>A to be uncertain at this time. - |
